Tirzepatide: Dual GIP and GLP-1 Receptor Agonist Research Overview

Compound Research
This page summarises findings from published academic literature. It does not describe or endorse use in humans or animals, and does not imply any treatment, prevention or cure of any condition.

Tirzepatide is a synthetic peptide studied in metabolic receptor pharmacology research as a dual receptor agonist. It acts on two incretin receptors, which is what sets it apart in the literature from single-receptor GLP-1 agonists. This page gives a general, research-framed overview of what published literature examines, for research reference purposes only.

What Is Tirzepatide?

Tirzepatide (development code LY3298176) is a 39 amino acid linear peptide. Its sequence is based on native GIP (glucose-dependent insulinotropic polypeptide), modified so that it also activates the GLP-1 receptor. A C20 fatty diacid is attached through a linker, which supports binding to albumin. Published pharmacokinetic studies report a half-life of around five days, much longer than the native hormones it is modelled on.

Areas of Published Research

Research on Tirzepatide sits within incretin biology, the study of gut-derived hormones and their receptors. Laboratory and preclinical work has examined its activity at the GIP and GLP-1 receptors, its signalling profile, and how dual receptor activation compares with activating either receptor alone. The molecule was first characterised by Coskun and colleagues in Molecular Metabolism in 2018.

Human clinical research was carried out by its developer through large phase 3 programmes, including SURPASS and SURMOUNT, which examined glycaemic and body weight endpoints. These programmes used products manufactured under medicinal controls and are referenced here for research context only.

Imbalanced and Biased Agonism

Tirzepatide does not act on its two receptors equally. Willard and colleagues (JCI Insight, 2020) described it as an imbalanced agonist, with greater relative activity at the GIP receptor than at the GLP-1 receptor. They also reported biased signalling at the GLP-1 receptor: it favours cAMP generation over beta-arrestin recruitment, which is linked in the literature to differences in receptor internalisation. This profile is one of the most studied aspects of the molecule.

Frequently asked questions

Is Tirzepatide approved for any medical use?

This page summarises research literature only. Pharmaceutical products developed from this molecule are separate products, manufactured and regulated as medicines. PerformanceLabsUK supplies research reference materials for in vitro laboratory use only. They are not medicines and are not for human or veterinary use.

How is Tirzepatide different from a GLP-1 agonist?

Single GLP-1 agonists are studied at one receptor. Tirzepatide is studied at two, the GIP and GLP-1 receptors, with greater relative activity at the GIP receptor.

Is Tirzepatide tested before dispatch?

Yes. Every batch is third party tested and issued with a batch-specific Certificate of Analysis before dispatch.

Is Tirzepatide intended for human or animal use?

No. All compounds discussed on this site are supplied strictly for in vitro laboratory research use only, and are not intended for human or animal administration.

Where can I find the Certificate of Analysis?

The batch-specific Certificate of Analysis is available on request from support@performancelabsuk.com with your order number, and sample Certificates of Analysis will be published in the PerformanceLabsUK COA Library as they become available.

Does this page constitute medical advice?

No. This page is provided for educational and research reference purposes only. It is not medical advice and does not describe or endorse use in humans or animals.
